1.5% AGAR (for bacteria plates)

 

1)  Make solution of SOB using the following recipe:

      

     Per 1 Liter:

            20 g Bacto-Tryptone

            5 g Bacto-Yeast Extract

            .58 g NaCl, anhydrous (10mM final)

            .1875 g KCl, anhydrous (2.5 mM final)

            2 g MgCl2·6H2O (10mM final)

            2.5 g MgSO4·7H2O (10mM final)

            980 ml ddH2O

 

2)  Add enough Bacto-Agar to result in a 1.5% agar solution (e.g., 15 g/1000ml).

 

3)  Autoclave.

 

4)  Let cool until warm/hot, but not scalding (about 50°C, able to rest against skin).

 

5)  Add 50 ug/ml Kanamycin.

 

6)  Pour plates completely covering the bottom.

 

7)  Store at 4°C.
