LB BROTH, LB-TETRACYCLINE, LB AGAR, LB-TET AGAR

(1 LITER)

LB BROTH

-10 g NaCl

-10 g Bacto-tryptone

-5 g Yeast Extract

-Add dH2O to final volume of 1 liter

-Adjust to pH 7.5 with 5 N NaOH (~200 ul)

-Autoclave 30 min liquid cycle.

LB-TET BROTH

-Make 1 liter LB broth

-Autoclave

-Cool to 55°C

-Add 12.5 mg of filter-sterilized tetracycline

-Store broth in a dark, cool place as tetracycline is light-sensitive

LB AGAR

-Make 1 Liter of LB broth

-Add 20 g Bacto-agar

-Autoclave

LB-TET AGAR

-Make 1 liter LB agar

-Autoclave

-Cool to 55°C

-Add 12.5 mg of filter-sterilized tetracycline.

-Pour plates and store in a dark, cool place.
